4. 제목: Advanced technologies for detection and molecular profiling of tumor-derived extracellular vesicles
xosomes are membrane-bound vesicles (50 – 200 nm in diameter) actively secreted by cancer cells. A growing number of studies show that exosomes may function as reliable surrogates of parental cells and with their abundance and structural stability, these vesicles can better reflect overall tumor burden, overcoming limitations of tumor heterogeneity and sampling bias. Rapid and accurate analysis of exosomes in clinical samples is, however, often hampered even in the research setting by three practical challenges: lengthy isolation procedures requiring ultracentrifugation to isolate exosomes, general unavailability of ultrasensitive assay systems to analyze large clinical cohorts for multiple markers, and specific cancer markers that separate tumor-derived exosomes from host cell-derived exosomes. Sensitive, standardized assays, optimized for these nanoscale vesicles, have yet to be developed to fully evaluate exosomes’ clinical potential. In this presentation, our current efforts to develop advanced sensing systems for high-throughput analysis of exosomal biomarkers will be discussed. In particular, the clinical reliability of recently-advanced nano-plasmonic sensor facilitating diagnosis of pancreatic malignancy and new approaches to develop plastic-based devices for point-of-care testing will also be introduced.
